TL RCI™: Methods in Transmitted Light

Vertical bright-ﬁeld illumination

Suitable for stained amplitude specimens with

suﬃcient contrast.

The light beams are deﬂected vertically through

the specimen. This results in an accurate bright

ﬁeld with maximum brightness.

Inclined transmitted light

Suitable for semitransparent, opaque speci-

mens such as foraminifera and ﬁsh eggs. Move

the deﬂection mirror until the desired data are

visible.

Single-sided dark ﬁeld

Suitable for ﬁxed specimens and ﬁne struc-

tures.

The ﬂatter the angle at which the light beams

are deﬂected into the specimen plane, the

darker the substrate appears. A dark ﬁeld-like

transmitted light is created. Outlines, ﬁne edges

and structures are bright, in contrast with the

dark background, through diﬀraction of the

light beams on the dark background.
